Abstract

Single nucleotide polymorphisms (SNPs) are highly abundant markers, which are broadly distributed in animal genomes. The variation in Myogenin (MyoG) and Myostatin (MSTN) gene will throw light on its potential use as a marker kit in selection. Hence, the study was conducted to detect single nucleotide polymorphismin fish Nile tilapia: (Oreochromis niloticus). Total 152 fish fin samples were collected from JangHang fish farm and divided according to their body weight and sex (Good male-35, Bad male-35, Middle male-12, Good female-35 and Bad female-35). Genomic DNA was extracted by using DNeasy Blood & Tissue Kit (QIAGEN). With the help of NCBI data, targeted regions like Promoter, 5’UTR and 3’UTR of each gene were amplified, electrophoresis, gel purification, sequenced, aligned, contiged and screened for SNPs. A total of 3 SNPs (from which one is Insertion TGACATATTTCCCTCACA) were located in 3’UTR of MyoG gene. In MSTN gene 5 SNPs and Short Tandem Repeats (STRs) were identified, from which one SNP, two Insertions and STRs(TG) in 5’UTR and two SNPs in 3’UTR. These collected information helps to get data for Genetic Marker kit.
